Beschreibung: Recognizes an 85-115 kDa protein (variation with cell type), identified as intercellular adhesion molecule (ICAM-1) (Workshop IV). It has 7 potential N-linked glycosylation sites. ICAM-1 is a single chain glycoprotein of Ig supergene family, present on unstimulated endothelial cells (EC) and on a variety of other cell types including activated fibroblasts, EC, macrophages, and lymphocytes. ICAM-1 mediates cell adhesion by binding to integrins CD11a/CD18 (leukocyte adhesion molecule, LFA-1) and to CD11b/CD18 (Mac-1). This interaction enhances antigen-specific T-cell activation. ICAM-1 also binds to CD43 and to Plasmodium falciparum infected RBCs. W-CAM-1 MAb blocks aggregation of cell lines mediated by the ICAM-1 and blocks homotypic binding of purified populations of activated T- and B-lymphocytes and also aggregation of mixed T- and B-cell blasts. It inhibits T-cell adhesion to normal human endothelial cells. Activation induced by cell-cell contact (mixed lymphocyte reaction, T-cell mediated B-cell activation) is significantly inhibited. This MAb blocks elements of both effector arms of immune system (cytotoxic cell function and Ig production).

Beschreibung: Recognizes a 67 kDa glycoprotein, which is identified as CD33 (HLDA IV; WS Code M-505). CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It belongs to the immuno-receptor tyrosine-based inhibitory motif (ITIM)-containing molecules able of recruiting protein tyrosine phosphatases SHP-1 and SHP-2 to signal assemblies; these ITIMs are also used for ubiquitin-mediated removal of the receptor from the cell surface. CD33 is expressed on cells of myelomonocytic lineage, binds sialic acid residues in N- and O-glycans on cell surfaces, and is a therapeutic target for acute myeloid leukemia. CD33 is expressed on myeloid progenitors, monocytes, granulocytes, dendritic cells and mast cells. It is absent on platelets, lymphocytes, erythrocytes and hematopoietic stem cells.

Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. Beschreibung: Thyroglobulin is a 660 kDa dimeric pre-protein with mutiple glycosylation sites. It is produced by and processed within the thyroid gland to produce the hormone thyroxine and triiodothyronine. Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation. The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for anti-thyroglobulin even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently anti-thyroglobulin negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. This antibody is useful in identification of thyroid carcinoma of the papillary and follicular types. Presence of thyroglobulin in metastatic lesions establishes the thyroid origin of tumor. Anti-thyroglobulin, combined with anti-calcitonin, can identify medullary carcinomas of the thyroid. Furthermore, anti-thyroglobulin, combined with anti-TTF1, can be a reliable marker to differentiate between primary thyroid and lung neoplasms.

Antibody to CK17 is an excellent tool to distinguish myoepithelial cells from luminal epithelium of various glands such as mammary, sweat and salivary. CK17 is expressed in epithelial cells of various origins, such as bronchial epithelial cells and skin appendages. It may be considered as anepithelial stem cellmarker because CK17 Ab marks basal cell differentiation. CK17 is expressed in SCLC much higher than in LADC. Eighty-five percent of the triple negative breast carcinomas immunoreact with basal cytokeratins including anti-CK17. Also important is that cases of triple negative breast carcinoma with expression of CK17 show an aggressive clinical course. The histologic differentiation of ampullary cancer, intestinal vs. pancreatobiliary, is very important for treatment. Usually anti-CK17 and anti-MUC1 immunoreactivity represents pancreatobiliary subtype whereas anti-MUC2 and anti-CDX-2 positivity defines intestinal subtype.

CK14 is found in basal cells of squamous epithelia, some glandular epithelia, myoepithelium, and mesothelial cells. Anti-CK14 is useful in differentiating squamous cell carcinomas from poorly differentiated epithelial tumors. Anti-CK14 is one of the specific basal markers for distinguishing between basal and non-basal subtypes of breast carcinomas. Anti-CK14 is also a good marker for differentiation of intraductal from invasive salivary duct carcinoma by the positive staining of basal cells surrounding the in-situ neoplasm as well as for differentiation of benign prostate from prostate carcinoma.
